    In protein engineering and antibody–drug conjugate (ADC) research, achieving “precise site-specific modification” of proteins has long been a critical scientific challenge. Traditional protein‑modification strategies typically rely on random reactions with the side chains of natural amino acids, leading to inherent uncertainties in modification sites, modification ratios, and product heterogeneity—factors that significantly impact both functional studies and drug development. In recent years, a biological strategy based on Genetic Code Expansion (GCE) has gained increasing attention. By reprogramming the protein translation machinery, this approach incorporates non‑natural amino acids at predefined codon positions during protein synthesis, enabling precise structural design of proteins at the expression stage. Unlike conventional methods that depend on post‑translational modifications, GCE shifts the control point upstream to the protein biosynthesis process itself, transforming protein modification from a stochastic event into a programmable, translational‑level process. This dramatically enhances the specificity and consistency of site‑directed modifications. This strategy offers a more precise and controllable biological platform for applications such as antibody–drug conjugation, protein labeling, and the design of functional proteins. In this live session, we will explore the principles and applications of site‑specific incorporation of non‑natural amino acids, highlighting their significance in ADC development and protein engineering.
