    Site-specific incorporation of non-canonical amino acids endows proteins with novel structures and functions.

    Non-natural amino acids (nnAAs)have structurally diverse side chain groups, and inserting them into proteins can endow them with new chemical properties, structures, and biological functions.

    PLD Technology relies on its independently developed Cell-free protein expression platform to achieve efficient and rapid synthesis of non natural amino acid inserted proteins. PLD Technology  has pioneered non-natural amino acid site-specific insertion services and has successfully completed multi-site nnAA site-specific insertion contracts on a few protein candidates of proteins, antibodies, carriers, etc.

    pAcF and pAzF targeted insertion

    p-acetyl-phenylalanine (pAcF) and para-azido-phenylalanine (pAzF) can be used as the coupling reaction site between antibody and drug to realize the precise control of drug-antibody ratio.

    Case Presentation

    Stable expression in a cell-free system makes membrane protein purification no longer challenging.

    PLD Technology  has developed its own CFPS system,where both nnAA and the according orthogonal translation systems are there to achieve site-specific insertion of non natural amino acids into protein sequences.

    Figure 1 shows the insertion efficiency of phenylalanine azide(pAzF) and phenylalanine acetyltransferase (pAcF) intomultiple sites of GFP. GFP with nnAAs at multiple sites stillshowed high expression levels;

    Figure 2 shows the mass spectrometry analysis results ofwild-type GFP and GFP with pAcF inserted at position 133. Asshown in the figure, the correct insertion rate is extremelyhigh.
