In protein engineering and antibody–drug conjugate (ADC) research, achieving “precise site-specific modification” of proteins has long been a critical scientific challenge. Conventional protein‑modification strategies typically rely on random reactions with the side chains of native amino acids, leading to inherent uncertainties in terms of modification sites, modification ratios, and product heterogeneity—factors that significantly impact both functional studies of proteins and drug development.
In recent years, a biological strategy based on Genetic Code Expansion (GCE) has garnered increasing attention. This approach reprograms the protein translation machinery to incorporate non‑natural amino acids at predefined codon sites during protein synthesis, thereby enabling precise control over protein structure at the expression stage. Unlike conventional methods that rely on post‑translational modifications, this strategy shifts the regulatory checkpoint upstream to the protein biosynthesis process itself, transforming protein modification from a stochastic event into a programmable process at the translational level. As a result, it markedly enhances both the site specificity and consistency of such modifications.
This strategy offers a more precise and controllable biological toolkit for antibody–drug conjugation, protein labeling, and the design of functional proteins.
